Cell Line Information: Cell Line Name: Malme-3M (ATCC® HTB-64™) Organism: Homo sapiens, human Tissue Derived from metastatic site: lung Cell Type: fibroblast Morphology: mixed Culture Properties: mixed adherent-suspension Biosafety Level; 1 Disease: Malignant melanoma Age: 43 years Gender: male Ethnicity: Caucasian Applications: This melanoma cell line was isolated from the same patient as Malme-3 (ATCC HTB-102), a normal skin fibroblast. The two cell lines provide tumor and normal counterparts for comparative in vitro melanoma studies. This is one of an extensive series of human tumor lines isolated and characterized by J. Fogh. Markers: HLA A2, Aw30, B13, B40( /-), DRw7 Tumorigenic: Yes Malme-3M is one of the cell lines of the NCI-60 panel which represents different cancer types and has been widely utilized for drug screening and molecular target identification.The Transfection Reagent: Transfection of suspension cells, or suspension/adherent mixed cells, such as Malme-3M cell, has always been challenging, both technically and financially. The most common systems currently used for molecular delivery to these cells include expensive electroporation instruments and relatively toxic chemical formulations.Malme-3M Cell Avalanche® Transfection Reagent is a new class of unique chemical formulations specifically formulated and optimized for transfecting Malme-3M cells. Malme-3M Cell Avalanche® Transfection Reagent (human melanoma cell) can also be used on the following cells with high transfection efficiencies. LOX-IMVI CellM14 Cell SK-MEL-2 Cell SK-MEL-5 CellUACC-257 CellUACC-62 CellVero Cell 293 Cell 293T/17 Cell Recommended protocols for these cells will be provided with the reagent. The protocols usually provide satisfactory transfection efficiency with invisible cytotoxicity. However, optimization may be needed for certain type of cells. Optimizations may include: the amount of DNA and this transfection reagent; cell density; transfection reagent/DNA ratio, or incubation time for the mixture of transfection reagent/DNA etc. For best transfection result, we recommend using the respective cell type/cell line specific Avalanche transfection reagents. Those reagents have been optimized on both recipes and protocols, and have been proved to have the best transfection results for the respective cell lines or primary cells.
