A GREAT INTRO TO ELECTROPHORESIS AND MOLECULAR SEPARATION.
In this miniPCR Learning Lab, students identify themoleculesfound in a mixture of dyes using two molecular separation techniques. Students will learn the key principles of gel electrophoresis and paper chromatography, and compare the two techniques. This lab introduces students to key concepts about molecules and molecular properties that are relevant to future discussions of important biological macromolecules such as DNA and proteins.
Students will explore how physicochemical properties (mainly size and charge) of molecules influence their migration through an agarose gel or in a paper chromatography separation. They will make predictions about the molecular composition of unknown dye mixtures based onempiricalresults. These experiments are highly visual andlend themselves to engagestudents ofdiverse learning styles.
This Learning Lab is suitable for students from middle school to AP Biology, and it is aligned with National Standards. It utilizes the following molecular genetic techniques and analyses:
- Micropipetting
- Gel electrophoresis
- Paper chromatography
- Use of charge/size ratios in the analysis of macromolecules
- Use observation to inform predictions about unknown molecular samples
The blueGel™Electrophoresis Rainbow Lab kit contains reagents for 8 lab groups of 4 students each (32 students):
- 5 dyes of different molecular characteristics
- Chromatography strips and accessories for chromatography
- Contrast-enhancing paper for easier visualization of results
- Reagents are compatible with blueGel™ or other standard horizontal electrophoresis systems
